Glp3 vs GLP-1

Searching this expecting a comparison will not produce one, because only one of the two exists. GLP-1 is a hormone with seven medications targeting it. GLP-3 is not a hormone, not a drug class, and not a thing under development.
The belief is easy to arrive at honestly. Almost everything else in medicine that carries a number does escalate — drug generations, treatment lines, device versions. Applied to a hormone family it simply does not hold: the number records position on the precursor protein, and there are only two glucagon-like fragments on it.
What proglucagon actually produces
GLP-1 and GLP-2 are both cut from the same precursor protein. This is the complete list of what that protein yields.
- GlucagonRaises blood glucose. The hormone the whole family is named after.
- GLP-1Glucagon-like peptide-1. Regulates appetite and caloric intake; the target of every medication on this site.
- GLP-2Glucagon-like peptide-2. Acts on the intestine — increases intestinal blood flow and inhibits gastric acid secretion. Nothing to do with appetite.
- OxyntomodulinAlso cleaved from proglucagon. No approved analogue on the US market.
There is no GLP-3. The numbering stops at two because only two glucagon-like peptides are produced. No such hormone appears in the physiology literature, and a search of the FDA drug label database returns zero results for it.
Where the name does show up is in marketing. A product described as GLP-3, or as a next-generation successor to GLP-1, is using a name with no regulatory or scientific meaning. That is worth treating as information about the seller rather than about the product.

Common questions
- What comes after GLP-1 then?
- Not a higher number — additional targets. Tirzepatide adds GIP to GLP-1. Other combinations under investigation add glucagon. The naming reflects which hormones are targeted, not a version sequence.
- Is GLP-2 the "next one" instead?
- GLP-2 exists but does something unrelated: it acts on the intestine and its approved analogue treats short bowel syndrome. It is not a stronger GLP-1.
